Clinical Significance:
p63 consists of at least two isoforms, TAp63 and ΔNp63. The ΔNp63 isoform (p40) is predominant in basal and myoepithelial cells, as well as squamous cell carcinomas of lung and other sites. The 4A4 anti-p63 clone that has been in use for many years is actually a “pan-p63” antibody, identifying both the ΔNp63 and the TAp63 isoforms. As the TAp63 isoform has a much wider tissue distribution than p40, antibodies to p40 can serve as a more squamous-specific marker, e.g., in the context of lung carcinomas, than the 4A4 "pan p63" antibody.
